Peptide blends put two or more research peptides into a single lyophilized vial. They save preparation time when a study uses compounds together, but they also make documentation harder. A single-peptide COA answers one identity question. A blend's COA has to answer one for every component.

Common research blends

How blends are made

Each peptide is synthesized and purified on its own first. The purified peptides are then combined in set ratios, dissolved, filled into vials and freeze-dried together. The ratio, usually given as the milligrams of each component per vial, should be stated on the label and product page.

Why blend testing is different

Purity

For a single peptide, HPLC purity is the main peak's share of the total. In a blend there are several intended peaks, one for each component. A blend's purity figure usually reflects the combined area of all the intended peaks, so it doesn't tell you whether each component is present in the right amount.

Identity

Mass spectrometry should confirm the molecular weight of every component. A report that identifies only one peptide in a three- or four-peptide blend leaves the others unverified. Learn more in HPLC vs. LC-MS.

Ratio

The most thorough reports also measure the amount of each component, which confirms the ratio on the label. If ratio data is important to your work, ask the supplier whether it's available for your batch.

Storage and reconstitution

A blend is only as stable as its least stable component, so handle the vial according to the most sensitive peptide in it. General guidance:

  • Store lyophilized vials refrigerated or frozen, away from light and moisture.
  • Let the vial reach room temperature before opening it, which limits condensation.
  • Reconstitute with an appropriate diluent, such as bacteriostatic water, added slowly down the side of the vial. Swirl gently rather than shaking.
  • Keep reconstituted vials refrigerated and avoid repeated freeze-thaw cycles.

Our storage and handling guide and bacteriostatic water primer cover these steps in more detail.

Checking a blend before you use it

  1. Confirm the batch number on the vial matches the COA.
  2. Check that mass spectrometry identifies every listed component.
  3. Note the purity figure, and whether ratio data is included.
  4. Confirm the testing lab is named and independent.
